Time-course of ROS production in mouse pancreatic acinar cells. Stimulation of cells with 50 mM ethanol (EtOH) led to a significant increase in ROS generation over the basal (resting) value (full circles). In the presence of the antioxidant cinnamtannin B-1 (10 μM) alcohol-evoked response was significantly decreased (open circles). Stimuli were added to the cuvette at the time points indicated by arrows. At the end of the experiment 100 μM H2O2 was added to the cells as a control to show increased fluorescence following oxidation of the probe by H2O2. Graphics are representative of 6-8 such experiments.
