AmpA does not change the amount of total actin protein in the cells.A) Polyacrylamide gels were loaded with total protein from 1 x105 cells per lane and stained with Coomassie. B)The amount of protein loaded was determined to give a linear rangesignal when scanned on a densitometer. To control for protein loadingdifferences the ratio of intensity of the F-actin band to a referenceband was determined. C) Western blot of total protein using ananti-actin antibody. 1 x 105 cells were loaded per lane. Incontrol experiments this amount of protein gave antibody staining foractin which was linearly dependent on the amount of protein loaded.
