The heterogenous cell population can be sorted by the ability of cells of the endothelial lineage to take up ac-LDL 488 via specialized scavenger receptors. (a) Cells were incubated with 2.5 μg/ml ac-LDL-Alexa Fluor® 488 for 4 h at 37°C and 5% CO2. Cells were subsequently washed twice with PBS and successful uptake was visualized by fluorescence microscopy (left: brightfield, right: fluorescence). The scale bar depicts 100 μm. (b) By using cell sorting technology, cells that have taken up the acetylated LDL-AF488 conjugate were successfully separated from the remaining population with a purity of 92%. Cell distributions are depicted in the plots.
