Effect of IRE1 inhibitor on mutant proinsulin degradation. Insulin 2 C96Y-GFP cells were treated with Dox for 24 h then treated or not with 100 μM cycloheximide (CHX) with or without p97/VCP inhibitor DBeQ (A) or 4μ8c (B). At the times indicated the cells were lysed and proteins were resolved by SDS-PAGE and immunoblotted with anti-GFP and GM130 antibodies. C. Western blot results were quantified and normalized to t = 0 h of cycloheximide treatment (Mean ± SE of 4 independent experiments). D. Analysis of Herp protein expression by western blot analysis.
