Treatment of cells with actin interfering drugs. HeLa cells were treated for 30 min at 37°C with the actin-binding drugs LatA (1μM), CD (1μM) Jaspl (200 nM), or vehicle (DMSO) and then stained with Hoechst 33342. Nuclear shape phenotypes were distinguished ("normal/round" or "irregular" nuclei) and counted by widefield microscopy. (A) The bar graph shows the percentage of normal nuclei (white) versus irregular nuclei (grey) measured from two independent experiments (see methods).
