Knockdown of PKCĪ“ inhibits osteogenic differentiation in hBMSCs. The knockdown efficacy of PKCĪ“ shRNA was assessed by RT-PCR (A) and western blot analysis (B). (C) Control shRNA- or PKCĪ“ shRNA-transduced cells were cultured in GM or ODM. ALP activity assay was determined after osteogenic induction of hBMSCs for 7 days. (D) For quantitative determination, the absorbance at 405 nm of ALP reaction was measured and expressed as the fold change of cultured cells over control shRNA-transduced cells in GM. (E) Control shRNA- or PKCĪ“ shRNA-transduced cells were cultured in GM or ODM, and differentiated osteoblasts were stained with alizarin red S. (F) For quantitative determination, the absorbance of alizarin red S was measured at 570 nm and expressed as the fold change of cultured cells over control shRNA-transduced cells in GM. (G) The mRNA expression of ALP, RUNX2, and OCN was estimated by RT-PCR. The representative images from three independent experiments are shown. Data shown are means Ā± S.E.
