Suppression of PKCĪ“-mediated osteogenic differentiation enhances the adipogenic phenotype of hBMSCs. Post-confluent hBMSCs were cultured in ODM with 2 Ī¼M rottlerin (A) or 10 Ī¼M compound C (G). (D) Control shRNA- or PKCĪ“ shRNA-transduced cells were cultured in ODM. Differentiated adipocytes were stained with oil red O after 14 days of osteogenic induction and at magnification of x 100 photographed. (B, E, and H) After 14 days of osteogenic induction, RT-PCR was performed to estimate the mRNA expression of adipocyte-specific genes, including PPARĪ³, C/EBPĪ±, and aP2, in the indicated groups. (C, F, and I) After 14 days of osteogenic induction, the protein expression levels of PPARĪ³ and C/EBPĪ± were determined by western blot analysis with the specified antibodies in the indicated groups.
