Reduced primitive endoderm differentiation of ES cells deficient of lamin A/C and/or emerin. a A Western blot shows the absence of lamin A/C and/or emerin proteins in ES cell lines with lmna (−/−) and/or emd (−/−) genotypes. Cells were incubated for 4 days with 1 μM RA, and cell lysates were prepared. b Wild type, lmna (−/−) and/or emd (−/−) ES cells were treated with or without RA for 4 days. The cell lysates were analyzed by Western blot. Protein loading was normalized to total (pan) ERK1/2. c The undifferentiated and differentiated cells were analyzed by qRT-PCR for expression of various markers. Values for each sample were normalized to respective GAPDH levels and plotted relative to control, untreated wild type (wt), which is defined as “1”
