Times for at least six and up to 21 cells for each copine regardless of the location of the GFP were averaged. The percent difference between the mean fluorescence intensity on the plasma membrane and the cytosol was calculated using images taken before cAMP stimulation and images in which the most membrane localization was observed after cAMP stimulation.
