RORα response elements (ROREs) of the NCEH1 promoter are directly activated by RORα. a HEK293 cells were transiently transfected with the luciferase reporter plasmid in the presence of pSV-βgal. As a luciferase reporter plasmid, wild-type (wt) or mutant (mt) ROREs with three direct repeats, as found in the NCEH1 promoter region, were used. At 36 h after transfection, HEK293 cells were co-transfected without (pSG5) or with RORα expression vector (pRORα). Luciferase activity in the cell lysates was determined and expressed as fold-change in RORα activation based on luciferase activity.
