Cell invasion assay. DU145 cells were seeded in matrigel-coated transwell chambers. 5 × 105 DU145 cells were transfected by pAAV-sFLT01-HisTag-GFP and/or control (pAAV-MCS-GFP transfected DU145 cells). Serum-free media was prepared and added to the matrigel-coated transwell chambers. The chambers were then placed in the lower plate which had been filled with 10% FBS containing media, After 24 h the non-invading cells were removed, membranes were fixed in methanol, and stained with DAPI. a Dapi staning of DU145 cells on insert (without matrigel), (B) Dapi staning of DU145 cells on insert coated with matrigel. c Dapi staning of DU145 cells which had been transfected by pAAV-MCS-GFP, as a control. d Dapi staning of DU145 cells which had been transfected by pAAV-MCS-GFP, on insert coated by matrigel as control. e Dapi staning of DU145 cells which had been transfected by pAAV-sFLT01-HisTag-GFP, on insert. f Dapi staning of DU145 cells which had been transfected by pAAV-sFLT01-HisTag-GFP, on insert coated matrigel.
