Mutation of lysine residue 917 decreases the ubiquitination of PDGFRα after short-term ligand stimulation. PAE cells transfected with either HA-tagged WT or K917R mutant PDGFRα (a), or tet-inducible PAE-PDGFRα cells induced with doxycycline to overexpress WT or K917R mutant PDGFRα (b), were serum-starved for 24 h and then stimulated with PDGF-AA for indicated time periods. An antibody against HA or PDGFRα was used to immunoprecipitate PDGFRα, followed by immunoblotting with indicated antibodies. Total cell lysates were also subjected to immunoblotting with antibodies against HA, PDGFRα, and α-tubulin which was used as a loading control. Ub, ubiquitin.
