From: Dexamethasone in osteogenic medium strongly induces adipocyte differentiation of mouse bone marrow stromal cells and increases osteoblast differentiation

Dex addition in standard osteogenic medium induces adipocyte marker expression. Relative levels of mRNA adipocyte markers (PPARgamma, glut4 (glucose transporter type 4), adiponectin (AdipoQ) and leptin) were determined on BMSCs cultured for 14 days in growth medium (G 0), in growth medium added with Dex 100 nM (G 100), in adipogenic medium containing Dex 100 nM (A 100) or 500 nM (A 500), or cultured in osteogenic medium (O 0) or osteogenic medium supplemented with 75 to 150 nM of Dex (O 75 to O 150). Levels of adipocyte marker mRNAs were reported to the mean of mRNA level of two housekeeping genes Gapdh2, and 18S and expressed in percent of cells cultured in growth medium. Representative data from four independent experiments depicted as means +/− S.E.M. are shown. # p < 0.05 vs. A 500 medium.
