Lamin A/C- and/or emerin-deficient ES cells show gene expression defects during differentiation. Wild type (wt), lmna (−/−) and/or emd (−/−) ES cells were treated with or without RA, and analyzed by confocal immunofluorescence microscopy for Oct3/4 and Gata4. a Representative examples of ES cells treated with or without retinoic acid are shown. b Oct3/4 and Gata4 expression was quantitated in the ES cells treated with 1 μM RA for 0, 3, 6, and 9 days. About 200 nuclei in each group were scored for Gata4 and Oct-3/4 staining, expressed as the percentage of the total number of nuclei.
